消化器癌の発症・進展過程における新しい遺伝子変異導入機構の解明

Research Abstract

消化器癌の炎症発癌における遺伝子変異導入機構を検討した結果、以下の結果を得た。
1) ヒトH. pylori感染胃粘膜、HCV感染肝組織、さらに潰瘍性大腸炎粘膜において、遺伝子編集酵素である種々のAPOBECファミリーの発現を見たところ、AIDのみの発現が見られ、他の分子の発現は認められなかった。またその際AIDの発現はNFκBの活性化と平行していた。
2) ヒト癌細胞を用いたIn vitroの実験では、AIDの発現はTNFα、IL4, IL13, TGFβによって増強し、その際、それぞれNFκB, GATA6, Smad4の活性化を介していた。
3) またヒト胃癌細胞ではH. pyloriが、肝細胞ではHCV core蛋白がAID発現を増強したが、その際NFκBシグナルをブロックすると、いずれによるAID発現も抑制された。なおH. pyloriについては、CagPAI-KOヘリコではAID発現はまったく見られず、一方CagA-KOヘリコではその活性は30%低下した。したがってH. pyloriは、4型分泌装置を介して何らかの因子を細胞内に導入してNFκBを活性化しAID発現を誘導するものと考えられた。
4) こうしたH. pylori,HCV刺激によって、P53をはじめとする様々な遺伝子変異が発生したが、本遺伝子変異はAID siRNAを導入することによって有意に抑制された。したがって、H. pylori, HCVによる遺伝子変異導入にはAIDが関与しているものと考えられた。なおこれらの変異の50%以上はnon-synonymous変異であり、また50%以上は、ヒトの癌にみられる変異と同一であった。
5) 以上より、H. pyloriやHCVによる炎症発癌では、AIDが発現して遺伝子変異蓄積に重要な役割を果たしているものと考えられた。
