| Research Abstract |
病原微生物を分離する生体材料として、血清を液体窒素中で凍結保存し、ヒト肝細胞キメラマウス接種し、2、7,14,21日と経時的に採血を行った。投与前の血清と投与後の各時点の血清を用いてrepresentational difference analysisにより投与前には存在せず、投与後に存在する核酸を分離同定し、塩基配列を確認することにより未知の病原微生物を検出することを試みている。
(1)これまでに液体窒素中に保存されている検体に加え、あらたに劇症肝炎、virus associated hemophagocytic syndromeの症例の血清を保存した。
(2)10症例分の検体を一群として、キメラマウスに経時的に投与した。投与前、および投与後1、2,4,8,16週と血清を採取し、保存した。
(3)各血清から核酸を抽出し、adopter primerを用いて逆転写反応を行った。さらに、linker ligationを行い、linkerのcomplementary strandとなるprimerを合成、2nd strandの合成を行いdouble strand DNAを得た。投与後の検体から得られたDNAをtester、投与前の血清から得られたDNAを driverとしてrepresentational difference analysisを行い、病原体特異的な核酸を増幅した。
現在いくつかのバンドを切り出して、塩基配列の決定を行っている。
