タバコの喫煙と皮膚老化

Research Abstract

環境要因でしわの形成に関与する因子として、紫外線が重要な因子であるが、喫煙も重要な環境因子であることが、私たちの調査や他の大規模調査によって明らかとなっている。私たちの研究室では、本邦における喫煙と皮膚の老化、つまりしわ形成と関与するかを再度調査し、正常人培養線維芽細胞を用いることで喫煙による皮膚老化の機序に関して解析を進めてきた。タバコ煙には約3700の物質が含まれ、水溶性物質はその約半分ほどであり、タバコ煙に含まれる代表的なベンツピレンなどは、脂溶性のものである。ヘキサンによるタバコ煙の脂溶性分画の抽出を終わり、線維芽細胞での解析で3-methoxy-4-nitroflavoneなどのAhRの特異的阻害薬によってAhRを介してMMP-1の発現がみられることを明らかにした。さらに、ケラチノサイトをレンチウイルスshRNAを用いた系AhR knockdownして、ケラチノサイトにおけるタバコ煙抽出液(水溶性、ヘキサン)とAhRの働きの解析において、ヘキサン分画では、AhRを介した経路がその後の遺伝子発現に重要であることが明らかとなった。動物実験では、ヘアレスマウスを用い、タバコ煙抽出液を外用、局所注射、腹腔内投与などを行い、その皮膚の変化を経時的に観察し組織学的変化の検討を行った。外用、局所注射で明らかに真皮コラーゲンの減少が得られることが明らかとなった。
