放射線適応応答を応用した新放射線治療法の開発

Research Abstract

腸管のクリプト細胞のマイクロコロニーアッセイを用いて放射線適応応答の有無を調べた。ICRマウスを用いpriming doseとして0.05Gy、0.5Gy、1.0Gyを用いた。Priming dose投与後、6、12、24時間後にtest doseとして12Gyを照射した。Test dose投与後3.5日目にマウスを屠殺し腸管を採取、パラフィン切片を作成し、空腸全周あたりクリプトマイクロコロニー数をカウントした。0Gyコントロールでは57±6.7(平均値±SD)で、priming doseのみでは、0.05Gy、0.5Gy、1.0Gyそれぞれで、59±2.5、63±2.2、66±5.5で有意差を認めなかった。12Gyのみでは9.8±1.5で有意に減少するが、priming dose 0.5Gyを照射し、12、24時間後に12Gyのtest doseを照射した群のマイクロコロニー数はそれぞれ15±5.4、17±3.0で、増加傾向を認め、また1.0Gy照射後12時間後12Gy照射したで群では23±6.5とクリプト数が有意に増加した(p=0.05)。腸管クリプト細飽で放射線適応応答が存在すると思われる。
なお、priming dose 0.05Gy群ではクリプト数の増加は認めなかった。
