Research Abstract

平成19年度に引き続き各種の光感受性物質の光線力学的治療効果について検討した。昨年度はアクリジンオレンジ(以下AO)と化学構造式がきわめて類似したメチレンブルーを中心に研究を行なってきたが本年度は同じ手法を用いてAzure A/B/C、Nile Blue、Toluidine Blue、Trypan Blue、Nile Red、Methylene Green、Rose Bengal、Congo Red、Methyl Violet、Alcian Blue、Safranin O、Rhodamin 123などの各種色素およびδ-aminolevulinic acid、ProtoporphyrinIX、Photofrin、Haematoporfhyrin、Verteporfinなどのポルフィリン系誘導体、Riboflavin、Acriflavin、Vitamin D3などのビタミン剤について殺細胞効果を検討した。なお対象細胞はマウス骨肉腫細胞株のLM8を用いた。各種色素では光線力学的治療効果が得られる濃度では細胞毒性も高度でAOのような光感受性による特異的な殺細胞効果は得られなかった。また腫瘍特異的な集積は認めなかった。ポルフィリン系誘導体およびフラビン体は効果を認めたがAOよりは弱く、蛍光も弱かった。以上より最終的にはAOが最も有効な光線力学的治療薬剤であると結論した。これらの研究成果については以下の論文で公表した。
