Outline of Final Research Achievements

本研究は、神経根への損傷が引き起こす脊髄内での細胞外環境の改変に着目したもので、神経の病理的・生理的両面の可塑性に関与している蛋白分解系である、組織型plasminogen activator(tPA)の活性と神経根症の引き起こす疼痛との関係を解明することを目的としている。以下の新規所見を見いだした。
1. ラットのL5神経根損傷モデルを作成し、脊髄後角でのtPA発現の経時的な変化を確認しその変化を定量した結果、術後3日目をピークに7日目まで有意に増加していた。
2. 脊髄後角でのtPA発現と、各種のマーカー蛋白とtPAを組み合わせた多重染色を行った結果、脊髄後角のアストロサイトがtPAを発現していることが明らかとなった。
3. ラット脊髄後根損傷による脊髄後角のプロテアーゼ活性の変化をIn situ Zymographyを用いて検討した。後根損傷後の脊髄表層において、プロテアーゼ活性の増加が確認され、tPA-STOPを投与するとその変化が消えることより、この脊髄後角におけるプロテアーゼ活性の増加はtPAの活性増加を介していることが明らかとなった。
4. 疼痛行動を起こす脊髄根部分損傷モデルにおいてtPAの発現を検討した結果、アストロサイトでのtPA発現は14日後でも増加していた。後根の部分損傷モデルにおける足底刺激に対する過敏反応は、tPAの特異的阻害剤のtPA-STOPの髄腔内投与によって、有意に抑制されることがわかった。
以上の結果は、英文雑誌GLIAに採択され、発表した。
その後、tPAのプロテアーゼ活性の標的蛋白を見つけるために、laminin, fibronectin, NR-1, BDNFなどを検索してきたが、現時点ではtPAとの新規メカニズムは見つかっていない。
