Research Abstract

本研究の目的は、我々がこれまでに発案したダイナミックマイクロアレイを利用して、細胞の網羅的な解析が可能なマイクロ流体システムを実現することである。
一万個レベルの細胞のアレイ化を実現するために、まずは、細胞サイズのビーズのアレイ化を検討した。さらに取り出しに細胞へのダメージを軽減させるためのデザインを検討した。これまでには、レーザの照射によりバブルを発生させていたが、熱に拠るダメージが危惧されてきた。そのため、バブル発生源をサンプルから遠ざける機構を検討した。
また、ここでは、我々が考案したプロセスを用いて、アルギン酸カルシウムゲルによるマイクロビーズを作成し、サイズ調整、細胞の分散化などの条件を導き出し、効率的なカプセル化を行なった。これによって、細胞を均一直径のゲルビーズで囲むことよって、細胞自身の直径のばらつきを軽減でき、効率的にアレイ化することに成功した。
