Research Abstract

Izumoはほ乳類の受精時に精子側で機能する膜融合因子である。Izumoの免疫グロブリン様ドメイン内に存在するN結合型糖鎖の機能を明らかにするために、非糖鎖型のミュータントIzumoを発現するマウスを作製した(N204QIzumo)。その結果、N204QIzumoの精子がほとんど精子-卵子の膜融合をしていないことが分かった。この原因は、精子の成熟過程で機能型Izumoが分解され、減少していることであった。このことから、IzumoのN結合型糖鎖はタンパク質の安定性に機能していることが明らかになった。
