Research Abstract

染色体末端に存在するテロメアの構造維持に重要なテロメレース逆転写酵素(telomerase reverse transcriptase: TERT)の新規機能の生理的意義とりわけ幹細胞の機能維持におけるTERT 発現の意義は、これまでに同定されてきたテロメレース複合体とは異なる機能構造体により維持されていることが強く示唆されることから、TERT がTERC 以外のRNA 結合パートナーとともに、従来知られてきたテロメア構造維持以外の新規機能を発揮するとの実験仮説のもと、TERT に結合するRNA の網羅的解析を行った。その結果、新規同定したRNA はこれまで、hTERTと安定な結合をするとされてきたRNA であるTERC と類縁のsmall nucleolar RNA snoRNA)に分類されるRnase mitochondrial RNA processing endoribonuclease (RMRP)であった。新規に同定したRMRP とTERT 複合体が、幹細胞やがん幹細胞機能維持において重要な役割を担うことが示唆された。
