Research Abstract

本課題の目標は,海馬の神経回路網の解析によって,精神疾患の新たな治療法である脳深部刺激法を導入するための基盤を確立することである.このために,電気痙攣刺激(ECS)を用いてうつ病治療モデルマウスを作成し,うつ病治療機序における海馬の神経回路網の構造と機能の変化について解析を行った.一連の研究から,ECSによって,海馬のミクログリアの突起密度が減少すること,GABA合成酵素であるGAD65とGAD67の発現様式が神経回路特異的に変化することを見出した.また,ECSによって,ミクログリアやアストロサイト,GABAニューロンの空間分布密度は変化しないことを明らかにした.これらの結果は,うつ病の治療機序において,組織学的細胞構築の変化を伴わない海馬の神経回路機能の可塑的な変化が主座にあることを示唆している.
