Research Abstract

本研究の目的は,ヒトTK6細胞のゲノム内で引き起こされる様々なDNA 付加体の突然変異誘発能を解析できる実験系を確立することである。そのために,まず代表的なDNA 付加体である8オキソグアニンを1つ含むターゲティングベクターの合成系を確立した。次に,それを特別な手法でTK6 細胞のゲノム内に部位特異的に導入させ,DNA 修復・複製させた後,そのゲノム内において8-オキソグアニンが起こした突然変異誘発パターンと頻度を解析することに成功した。現在,それらの実験系とデータの信憑性や再現性について調べている。
