Research Abstract

分子内水素結合による水酸基のpK_aシフトを利用して、脂肪族リン酸モノエステルの脱リン酸化を検出する新規スイッチ機構を考案し、その原理によるレシオ蛍光プローブを複数合成した。これらの化合物を含む100 mM HEPES buffer (pH 7.4) に、acid phosphatase (ACP)を加え励起スペクトルを測定したところ、励起スペクトル変化が観測された。種々のホスファターゼに対する選択性を調べた結果、全てのプローブが酸性ホスファターゼに対して高い選択性を持つことを明らかにした。
