Research Abstract

シロイヌナズナを用い,HSP90がどのような細胞内因子と結合し,その働きを調節するかを調べるため,HSP90阻害剤を処理したシロイヌナズナの応答を解析した.その結果,植物においてHSP90は熱ショック応答を制御していることと,オーキシン応答を制御していることがわかった.HSP90はシロイヌナズナにおいて常に細胞内に蓄積している熱ショック転写制御因子,HSFA1と結合し,この結合はHSP90阻害剤により阻害された.HSP90阻害剤処理によりHSFA1が活性化された.これらのことから,HSP90は熱ショック転写制御因子と結合しその活性を負に制御していることが示唆された.一方,オーキシンで誘導されるレポーター遺伝子をもつシロイヌナズナを用い,HSP90阻害剤で,オーキシン応答が顕著に抑制されることがわかった.さらに,HSP90阻害剤で,内在性のオーキシン応答遺伝子の発現が顕著に抑制された.これらのことから,HSP90はオーキシン応答を制御することが示唆された.
