Research Abstract

細胞分化等を誘導する分子の細胞内局在変化の引き金となるImportin ファミリーの一つ、Ipo13遺伝子の減数分裂期生殖細胞細胞分化における役割について研究を進めた。全長(long)のIpo13(L-Ipo13)は標的タンパク質カーゴ分子であり核内で活性を示すubiquitin-conjugating enzyme 9(UBC9)の核局在を誘導する。しかし、精巣特異的発現を示す、核移行領域欠損型のTestis specific(TS)-Ipo13はUBC9の核への局在化を誘導しなかった。さらに両者の共発現ではL-Ipo13のUBC9の輸送活性が阻害されることを観察した。また、UBC9が核局在を示さない条件下では、生殖細胞の減数分裂の進行が遅れる様子を観察した。以上の観察から、転写開始点の使い分けによって得られると推測されるTS-IPO13は、Ipo13遺伝子の全長産物であるL-IPO13の細胞質核間輸送活性を低下させ、UBC9などの標的カーゴ分子の核蓄積を阻害して、減数分裂期の生殖細胞の分化を制御していると結論づけた。
