| Research Abstract |
グリコーゲンや穀物デンプンのリン酸化に関与する酵素を探索した. 大腸菌抽出液から酵素精製を試みた. 複数のクロマトグラフィーで分離する段階で, グリコーゲンリン酸化酵素の活性は急激に減少することが認められた. そのため, 目的タンパク質の同定には至らなかった. 一方, イネのデンプンリン酸化酵素は1457残基からなる162 kDaのタンパク質であることを明らかにした. これは完全長cDNAデータベースに登録されている構造とは異なっていた. 大腸菌で本酵素を発現させたところ, グリコーゲンのリン酸化率が上昇し, 酵素活性を有することが確認された.
