Research Abstract

本研究は、食事性AhRリガンドであるフラボノイド類のうち、フラボン、フラボノール、フラバノン、カテキンの各サブクラスから数種の化合物を選定し、これらの化合物がAhRの活性化に関わる事象に対する影響と、投与形態がその体内動態に及ぼす影響を明らかにすることを目的とした。その結果、1.フラボン、フラボノール、フラバノンに属する化合物はAhRとアゴニストとの結合を競合的に阻害するのに対して、カテキンは阻害しないことが分かった。2.培養肝細胞においてダイオキシン類が誘導するAhRの核内移行に対しては、フラボンとフラボノールに属する化合物がこれを阻害した。3.(-)-エピガロカテキン-3-ガレート(EGCg)のみを投与したときの総EGCgが43nMであったのに対して、GTEを投与したときの総EGCgが427nMであった。同じ動物個体から得た血清を用いてマウス肝腫瘍由来Hepa-1c1c7細胞に処理したところ、4.2,3,7,8-四塩化ジベンゾ-p-ジオキシンによって誘導されるシトクロームP450 1A1の発現を有意に抑制した。
