Research Abstract

タンパク質の多くは無味であるが、甘味を持つタンパク質の存在が複数知られている。ネオクリンは甘味と味覚修飾活性(酸味を甘味に換える活性)を併せ持つユニークなタンパク質である。その分子表面に塩基性アミノ酸が集まっていることに注目し、塩基性ヒスチジン残基をアラニン残基に置換したネオクリン変異体を作成した結果、それらの残基が味覚修飾活性に深く関与していることが明らかになった。ネオクリンは舌上に存在する甘味受容体によって認識されていることから、受容体側を遺伝子学的に改変し、受容体のどの部位がネオクリンと相互作用しているかについても一定の知見を得た。
