Research Abstract

ウシ体細胞クローン胚に特異的な発現動態を示す遺伝子の特定と、その制御因子のDNA メチル化状態が胚発生にともないどのように変化するのかを検討した結果、胚の分化や胎盤機能に関連する遺伝子においてクローン胚に発現異常がみとめられ、これら異常を示す遺伝子の発現量は胚発育にともない発現が正常化する遺伝子と、発現異常が継続する遺伝子が存在することが明らかとなった。また、クローン胚のDNA メチル化割合は、胚盤胞期において高いものの、その後の胚発生にともなって正常化することが明らかとなった。本研究によりウシ体細胞クローン胚の遺伝子発現動態とその制御機構の一端が明らかとなった。
