Research Abstract

仮性狂犬病ウイルス(PRV)は動物に重篤な神経症状を引き起こすため、獣医学領域で極めて重要なヘルペスウイルスである。PRV の転写調節因子IE180 を発現するトランスジェニックマウス (TgIE180)は小脳形成不全を呈し、IE180が小脳の発生に影響を与えることが示唆されている。本研究では、TgIE180の解析から、ウイルスの感染という現象を伴わずIE180が単独で神経細胞やグリア細胞に変性・細胞死を誘発することを明らかにし、IE180 の発現によりヘルペスウイルス感染の病態が一部再現されることを示唆した。また、IE180が細胞の恒常性維持に重要な蛋白質分解経路に関連する宿主分子に作用し、異常蛋白質の蓄積を促進する可能性を示した。
