Research Abstract

本研究ではエンド-β-N-アセチルグルコサミニダーゼが持つ糖タンパク質の糖鎖をすげ替える能力を変異導入により高めることを目的とし、エンド-β-N-アセチルグルコサミニダーゼ-M(Endo-M)のN173A変異酵素と酵素反応中間体であるオキサゾリン合成基質を組み合わせることにより、効率的な糖鎖転移方法の開発に成功した。また、エンド-β-N-アセチルグルコサミニダーゼ-A(Endo-A)のN171G変異酵素を用いることで、糖タンパク質への糖鎖転移活性を10倍高めることに成功した。本変異酵素を利用することにより、糖ペプチド及び糖タンパク質への糖鎖の導入効率を高めることができた。
