Research Abstract

PUFA が欠乏すると、知能発達障害、皮膚障害など様々な病態、疾患を引き起こす。これらの中にはプロスタグランジンなど脂質性メディエーターの欠乏で説明できるものもあるが、脳神経系の発達等については、アラキドン酸代謝物だけでは説明できず、PUFA が他にも重要な機能を果たしていると考えられる。PUFA はリン脂質に多く含まれており、柔軟な膜を形成することから、特に膜タンパク質の機能発現に重要な役割を果たしていることが予想されている。しかしながら、実際にどのような分子がPUFA を豊富に含む膜環境を要求するか、これまでほとんど明らかになっていない。
本研究では線虫C.elegans を用いた網羅的RNAi スクリーニングにより、機能発現にPUFA を要求すると考えられる遺伝子を複数同定した。また。これらの細胞内局在を解析したところ、膜タンパク質と考えられる機能未知分子Xの細胞内局在がPUFA欠乏変異体において変化することを見出した。この局在変化はPUFA の添加によりレスキューされたことから、本分子の適切な局在維持にPUFA を含む膜環境が必要であると考えられる。これまでPUFA 欠乏状態によって引き起こされる膜環境の変化が、どのような分子に影響を与えるか不明であった。本研究により同定した分子は進化的に高度に保存された膜タンパク質であり、哺乳動物においても機能発現にPUFA を豊富に含む膜環境を要求すると考えられる。
