Research Abstract

腸管出血性大腸菌O157:H7由来のベロ毒素の糖鎖認識サブユニット(Stx1B)は、抗原提示細胞の主要組織適合遺伝子複合体(MHC)クラスIIに抗原由来ペプチドが提示されにくいために免疫原性が低いと考えられている。これを改善するためにStx1Bと、MHCクラスIIに提示されるOVAペプチドを共存させたリポソームを作製しマウスに対して経粘膜免疫を行いその効果を検討した。リポソームワクチンの経鼻免疫により、OVAペプチド内封リポソームは粘膜での分泌型IgAの産生を誘導した。またペプチドを内封せずに表面にStx1Bを結合させたリポソームワクチンでも分泌型IgA産生が誘導された。さらに、Stx1Bをリポソームに内封することで免疫原性を高めることが可能であった。MHCクラスIIに提示されるOVA由来のペプチドを内封してStx1Bを表面に修飾したリポソームに改良を加えることで、効果的なリポソームワクチンの開発が期待される。
