Research Abstract

真性赤血球増加症患者で見出されたJAK2 V617F変異体は、恒常的な活性化を示すことが報告されていたが、その詳細な活性化機構およびJAK2変異体の機能は不明であった。本研究では、JAK2の活性に重要なチロシン残基として613を、また新規リン酸化部位としてY913を同定した。また、JAK2V617F変異体発現細胞が異常な増殖を示し、ヌードマウスに投与すると、腫瘍を形成することを明らかにした。
