Research Abstract

Glyoxalase I(GLO I)は,解糖系で生じるカルボニルストレスの消去システムを担う主酵素であり,大腸がん,肺がん及び前立腺がんや,抗がん剤に耐性を示す培養がん細胞株で高発現がみられることが報告されていることから,新しいがん治療薬開発のターゲットとして非常に有望視されている.そこで本研究では,GLO Iを新規制がん剤開発の創薬ターゲットとし,in silico設計及び有機合成展開によって新規GLO I阻害剤リード化合物を創製することを目的とした.まず,in silicoで基質遷移状態をmimicすることが予測された4位=O,5位-OH flavonoid化合物のGLO I阻害効果についての構造活性相関解析を行い,その解析結果に基づいてGLO I阻害剤設計のpharmacophoreを構築して,新規骨格をもつGLO I阻害剤のin silicoスクリーニングを行った.それらについてGLO I酵素反応生成物S-D-Lactoylglutathioneの240 nmにおける吸光度測定によるin vitro assayを行い,GLO I阻害活性を評価した.得られたGLO I阻害剤候補化合物について有機合成展開を行い,GLO I阻害活性をin vitro assayで評価し,構造活性相関解析を行った.in silicoスクリーニング及びin vitro assayによって,新規GLO I阻害剤候補化合物としてTLSC702を得た.TLSC702は,既存のGSH誘導体型GLO I阻害剤S-p-bromobenzylglutathione (BBG)よりも高いGLO I阻害効果を示した.これについて有機合成展開を行った結果,in vitro assayでTLSC702と同等のGLO I阻害活性を示す化合物が数種得られた.本研究によって新規骨格を有するGLO I阻害剤候補化合物が得られたことは,新規制がん剤開発のためのリード化合物創製において大変意義深いものであると考えられる.
