Research Abstract

本研究により、小腸上皮細胞・肝細胞の頂端膜においてステロール類の吸収に働き、エゼチミブの薬効標的であるヒトNiemann-Pick C1-like 1(NPC1L1)がSREBP2/HNF4αによりコレステロール依存的な制御を受けること、脂肪酸を生理的リガンドとする核内受容体PPARα/NR1C1により正の転写調節を受けること、および、糖尿病時に機能上昇が見られるPGC1αがSREBP2/HNF4αおよびPPARαによるNPC1L1 転写活性を亢進することが明らかとなった。これらの知見は、相互に増悪因子となる高脂血症と糖尿病の病態生理の解明に向けた重要な一歩となることが期待される。
