Research Abstract

本研究の目的は、リノール酸、アラキドン酸、ドコサヘキサエン酸などの多価不飽和脂肪酸に由来する酸化脂質をターゲットとしてオーファンGPCRのリガンドスクリーニングを行い、受容体を同定することを通して新たな生理活性をもつ酸化脂質を発見することであった。そのために、以下のような実験を行った。‐候補となるオーファンGPCRの選択と安定発現細胞株の作製‐アミノ酸配列の相同性に基づいた系統樹解析から、脂溶性の物質をリガンドとする可能性が高いと予想されるオーファン受容体を10個選定した。定法により受容体を安定的に発現するCHO細胞を樹立した後、受容体のN末端に付与したFLAGタグを免疫染色し、フローサイトメトリーによる解析で受容体の発現量が高いクローンを各受容体につき数個ずつ選択した。‐酸化脂質ライブラリー作製‐リノール酸(C18:2)、α-リノレン酸(C18:3、n-3系列)、γ-リノレン酸(C18:3、n-6系列)、アラキドン酸(C20:4)、エイコサペンタエン酸(C20:5)、ドコサヘキサエン酸(C22:6)およびこれらを構成成分として持つリン脂質を基質としてin vitroで酸化反応を行い、酸化脂質ライブラリーを作製した。酸化反応として、二価金属イオン(Cu2+またはFe2+)を用いた反応もしくはラジカルイニシエーター試薬を用いた反応を行い、反応時間を変化させることにより様々な程度に脂質を酸化させ、薄層クロマトグラフィー分析などにより酸化条件の至適化を行った。‐リガンドスクリーニングアッセイ‐作製した受容体安定発現細胞および酸化脂質ライブラリーを用いて、細胞内カルシウム上昇反応に基づくリガンドスクリーニングアッセイを行ったが、本研究期間中に陽性反応を得るには至らなかった。
