Research Abstract

細胞骨格因子β-tubulin,特にβIII-tubulin(以下βIII)は神経細胞特異的である.しかし多く腫瘍ではβIIIの異所的発現が見られ,タキソール系抗がん剤に対し抵抗性を獲得する.本研究ではβIII遺伝子の発現制御因子RESTに注目し,異所的βIII発現機構,さらに薬剤耐性機構を検討した.その結果,RESTが細胞周期依存的に変動しβIIIを産生すること,また悪性黒色腫細胞でβIIIの発現が低いものは,タキソールに高感受性であることを明らかにし,一部の悪性黒色腫に対するタキソールの有効性を示唆した.
