Research Abstract

クロマチンリモデリング因子CHD(Chromodomain-Helicase-DNA binding domain protein)ファミリーに属する酵素群は、クロマチン構造の変化を介して遺伝子発現の制御に関わると考えられているが、特定遺伝子の発現制御における役割は不明であった。最近申請者は、このファミリーの一つであるCHD8が発生早期に高発現し、がん抑制タンパクp53によるアポトーシスを抑制することを発見した[Nishiyama M, et al., Nature Cell Biol.(2009)]。CHD8はp53とヒストンH1との三量体を形成することによって、p53の転写活性能を抑制し、最終的にアポトーシスを阻害する。つまりCHD8はヒストンH1を呼び込むことによってクロマチン構造を変化させ、p53による転写を抑制することによって、細胞運命をアポトーシスから生存へと180度変化させる。CHD8が欠損すると、p53が異常に活性化してアポトーシスが誘導される。CHD8ノックアウトマウスでは胚発生早期に広範なアポトーシスを来たし死亡するが[Nishiyama M, etal., Mol. Cell. Biol. (2004)]、p53を同時欠損させるとアポトーシスが抑制され、この発生停止が回復した。これらの生化学的・遺伝学的知見により、CHD8は既にクロマチン上に結合しているp53すらも抑制できる"抗p53最終機構"を形成していることが判明した
