Research Abstract

蛍光ラベルしたMycobacterium tuberculosis H37Rv(結核菌)を、GFP融合各種Rab GTPaseを発現するRawマクロファージ細胞に感染させて、Rabタンパク質が局在している結核菌ファゴソームの割合を共焦点レーザー顕微鏡で調べて、黄色ブドウ球菌ファゴソームと比較した。Rab8、Rab9、Rab22bなどは黄色ブドウ球菌、結核菌ファゴソームと同様の局在変化を示した。Rab11、Rab20、Rab37は黄色ブドウ球菌ファゴソームに局在するが、結核菌ファゴソームには局在しなかった。Rab23、Rab34、Rab39 はRab7と同様に結核菌ファゴソームに局在するが、その局在は黄色ブドウ球菌に比べて制限されていた。
結核菌ファゴソームからかい離するRab GTPase のファゴソーム熟成における機能を明らかにするために、これらのドミナントネガティブ変異体を発現するマクロファージにおいて、ファゴソームの酸性化とファゴソームへのカテプシンDの局在を調べた。Rab7、Rab20、Rab39はファゴソームの酸性化に、Rab7、Rab20、Rab32、Rab34、Rab38がカテプシンD のファゴソームへの局在に関与することが明らかになった。以上の結果は、ファゴソーム熟成とファゴリソソーム形成に数種類のRabタンパク質が関与すること、結核菌はこれらのRab タンパク質の細胞内局在を変調させることによって細胞内寄生を獲得していることを示唆する。
