Research Abstract

免疫不全症の遺伝子治療では、治療用のレトロウイルスベクターの組み込みによるがん遺伝子の活性化により、副作用(白血病)が発生した。そこで、ベクターの安全性の再評価のために、ベクターの組み込み性状を解析し、特有の組み込み指向性とそれに伴う標的宿主遺伝子の転写制御を明らかにした。さらに、白血病に関与したがん遺伝子の活性化の機序をベクター組み込みの観点から解析し、がん遺伝子近傍でのベクター組み込み特性およびその頻度を算出した。これらの結果は、より安全な遺伝子治療を考える上で重要である。
