Research Abstract

本研究では、ストレス反応性蛋白ペリオスチンの心臓弁膜症進展における役割を解析した。ペリオスチンはマウス胎生期より心臓弁特異的に発現し、ヒト正常弁では内皮細胞下でα-smooth muscle actinと共発現しており、病的弁では血管新生と共にその発現領域は約4倍に増加した。In vitroではペリオスチンは内皮細胞の血管新生能を有意に増強した。高脂肪食負荷により、野生型マウスでは大動脈弁の有意な肥厚、弁輪部の血管新生・線維化の増加を認めたが、ペリオスチン遺伝子欠損マウスではそれらの変性が軽減しており、ペリオスチンの弁変性作用が示された。
