Research Abstract

ポリグルミン病において、近年、封入体形成は細胞死に対し抑制的に働くと唱えられ、細胞傷害性のある構造の同定が望まれている。本研究ではFRET (fluorescence resonance energy transfer) 技術を応用して封入体形成に先行するポリグルタミン鎖オリゴマーを可視化し、ポリグルタミン鎖が、長さ依存性に方向性をもってオリゴマー形成することを明らかとした。
ポリグルタミン鎖を安定発現するSH-SY5Y 細胞株を神経分化誘導後にFRET 陽性細胞群、FRET 陰性細胞群、封入体形成群間の細胞生存期間解析をおこない、FRET 陽性細胞群において強い神経細胞死を認めた。これにより、ポリグルタミン鎖は、自己重合したオリゴマー状態で強い細胞毒性を獲得することを証明した。
