Research Abstract

本研究はアルツハイマー病の病的分子アミロイド及びグリア細胞の活性化の生体イメージング及び摘出脳の免疫染色により、アミロイドの産生・除去とグリア細胞の活性化との関連を調べた。その結果、ポジトロン断層撮影(PET)を主なる手法とする分子イメージングはアミロイド沈着と炎症性反応を臨床研究においても応用できることを明らかにし、グリア細胞活性化の指標である末梢性ペンゾジアゼピン受容体(PBR)は単なるグリア細胞のバイオマーカーでなく、グリア細胞の機能をも制御しうる機能性分子であることを見出した。特に中枢神経の免疫を担うグリアの亜種ミクログリアにおいて、PBR分子は非炎症性ミクログリア細胞が炎症性ミクログリアに変化する際に、その発現が高くなり、活性化ミクログリア細胞全般よりも、むしろ炎症性ミクログリアの挙動を特異的に反映するものと考えられる。また、PBR結合性薬剤はこれらの炎症性ミクログリアによる炎症性サイトカインの放出を制御しうることを明らかにした。
