Research Abstract

培養正常ヒト表皮角化細胞(NHK)をもちいてhornerinの発現条件を検討した。Ca2+添加による分化誘導ではProfilaggrinの発現はみられるものの、hornerinの発現がみられず、細胞をfull-confluentの状態に維持することによりhornerinの発現が確認された。さらにRNAiによりprofilaggrin発現の抑制した条件下でもfull-confluent を維持することによりhornerinの発現が誘導された。この結果よりhornerinとprofilaggrinは異なる発現調節を受けることが示唆された。また、各種角化異常を伴う皮膚疾患をもつ患者より得られた皮膚を用いてHornerinの発現を検討した。水疱型先天性魚鱗癬紅皮症や扁平苔癬、掌蹠角化症、慢性期アトピー性皮膚炎の一部などで発現が確認され、さらにhornerinが発現している皮膚ではサイトケラチン6の発現がみられた。これよりhornerinは過増殖状態にある表皮角化細胞で発現すると推測された。サイトケラチン6はTNF-αやIL-1など炎症性サイトカインの刺激で発現することが知られており、hornerinの発現にも炎症性サイトカインが関与関与しうるか現在器官培養系を用いて検討中である。以上の結果よりhornerinは表皮においてprofilaggrinと異なる機能を持つことが示唆され、さらにhornerinの発現は炎症性角化症などのヒト疾患の病態形成に関与していると考えられた。
