| Research Abstract |
PC12細胞にFluvoxamine(FLV)投与後、AKT-Ser473,Thr308のリン酸化が亢進していた。またBDNFをPC12細胞に投与後、同様にAKTのリン酸化が起こり、PI-3 kinazeの活性化を抑制するLY294002にて、BDNFによるAKTのリン酸化が抑制された。また同様にLY294002にて、フルボキサミンによるAKTのリン酸化が抑制されたことから、フルボキサミンによるAKTのリン酸化はBDNFを介して起っていると考えられた。FLV,Milnacipran(MIL)を3週間投与後の大脳皮質において、Akt-Ser473のリン酸化、AKT-Thr308のリン酸化が亢進していると考えられた。FLV、MILを3週間投与後、大脳皮質の核内でのAKTリン酸化の増加とリン酸化されたAKTの核への移行を認められた。核内に移行したAKTは転写因子をリン酸化し、転写因子とコファクターの結合を調節し、転写を制御していることが考えられた。
