Research Abstract

大腸癌においてDNAメチル化がどのように特定ゲノム領域に形成されるかを解明するため、癌遺伝子c-MYCの標的遺伝子とDNAメチル化形成との関連を検討した。ヒト大腸由来正常上皮細胞にc-MYCを過剰発現させると標的遺伝子の発現抑制をするもののDNAメチル化は誘発せず、当初の仮説を支持する結果は得られなかった。一方、新規に同定したc-MYC標的遺伝子ALEX1の大腸癌におけるDNAメチル化による不活化が観察された。大腸癌は多段階的異常の蓄積により進行すると考えられており、c-MYCの過剰発現に加え、その他遺伝子変異やシグナル伝達経路の異常活性化がDNAメチル化形成に必要であるのかも知れない。
