Research Abstract |
ラットのcranial window modelを用いることで、スフィンゴ脂質の一つであるsphingosylphosphorylcholine (SPC) がin vivoの実験系においてもRho-kinaseの活性化を介する脳血管攣縮を引き起こすことが判明した。また、高コレステロール食(F1飼料に1%コレステロールと1%コール酸を添加)で飼育した高脂血症ラットは、SPC刺激でコントロール群(F1飼料で飼育)よりも強い血管攣縮が認められ、SPCが引き起こす血管攣縮機構がコレステロールによって制御されている可能性も示唆された。本研究はくも膜下出血後の脳血管攣縮のメカニズムの解明につながるものである。コレステロールによる血管攣縮の制御機構の存在を示唆する結果は、脳血管攣縮発症の予測や予防的治療に強く貢献し、臨床的にも極めて大きな意義を有すると思われた。
