Research Abstract

誤嚥性肺炎は高齢者医療においては非常に重要な疾病である。簡易かつ効果的なインフェクションコントロール法の確立が求められており、我々は誤嚥性肺炎に対してプロバイオティクスを応用する事を目的としてマウスを用いた実験を行った。
5-10週齢のオスBalb/cマウスを用い、従来の研究で示されている誤嚥性肺炎モデルを応用した。この過程において死亡が確認されたマウスに関しては、肺組織内における膿瘍形成が認められ、肺炎が死因であることが考察された。術後一週間でマウス肺を摘出し、誤嚥性肺炎群、乳酸菌投与群、併用群、コントロール群(各5匹)に分けた。肺組織のtotal RNAを抽出してcDNAを作製し、サンプル中の遺伝子発現を定量的に解析し、炎症性サイトカインに関して検討を行った。結果、誤嚥性肺炎群において炎症性サイトカインの発現量が有意に高いことが明らかとなった。加えて、乳酸菌投与がその炎症反応を抑制する可能性が示された。
これらの炎症性サイトカインの濃度、局在に関してさらに検討を行い、免疫機構におけるプロバイオティクスの意義について解析する必要がある。本研究の結果は誤嚥性肺炎に苦しむ要介護者に対して非常に多くの利益をもたらし、歯科医学の発展に大きく貢献するものと予想される。
