Research Abstract

マウス骨細胞由来の細胞株MLO-A5を血清存在下で培養し、βグリセロリン酸およびアスコルビン酸を添加して石灰化を誘導した。単離した基質小胞を抗原としてSPFで飼育したラット(WKY/NCrj)の足部に皮下注射後、3週で腹部大動脈リンパ節を摘出し、B細胞を単離し、ミエローマ細胞をPEGで融合させ、約100万種類のハイブリドーマ細胞を作製した。この中から、基質小胞に特異的な物質を認識する抗体を産生するものを選択する目的で一次スクリーニングを行った。すなわち、対照として、同じくT抗原を遺伝子導入した遺伝子改変マウスの長幹骨由来の骨細胞より樹立した細胞株で、逆に基質小胞を分泌せず石灰化も生じないMLO-Y4細胞を使用し、ELIZA法を用いて、MLO-A5に結合するがMLO-Y4には結合しない有望なハイブリドーマ細胞を複数同定した。
