Research Abstract

回転分子モーターであるF1の反応力学モデルの構築を目指し、申請者が独自に開発した個々の触媒サブユニットへ特異的に変異を導入するハイブリッドF1の再構成手法を用いて、分子の内部状態の解釈まで可能にした1分子回転観察系を構築した。様々な変異体を用いたハイブリッドF1の回転について、新たに開発した角度解析プログラムを用いて解析を行った結果、これまで発見されていなかった新規回転ステップの素過程(約20度のサブ-サブステップ)が、各種変異体で普遍的に起こる事象であることが確認できた。この結果は、F1という回転分子モーターの回転メカニズムにとっての新たな発見となるだけでなく、生体分子機械がいかにして反応を力学運動に変換しているかを解き明かす鍵となる成果である。
