Research Abstract

本研究により、EMTに際してMSX2と共に誘導されるカルシウム結合蛋白S100Pが同定され、その誘導はMSX2と同様Smad4依存性であった。MSX2、S100Pの誘導にはいずれも新規の蛋白合成が必要であり、Smad4の下流の標的遺伝子がこの過程に重要であると推測された。ホメオボックス遺伝子であるMSX2が膵癌において癌幹細胞の性質のひとつである抗癌剤耐性を誘導し、その過程にはトランスポーター遺伝子であるABCG2の誘導が関わっていることが明らかにされた。プロモーター解析によりMSX2はABCG2の発現を直接制御していることが判明した。これらの分子機構は膵癌の新規治療標的となりうるものであると考えられる。
