Outline of Research at the Start

本研究は、①新規ナノ触媒である5酸化バナジウムナノ粒子(V2O5 NP)を数千個包埋した高分子ナノ小胞を作製し、抗体を修飾する。②異物や血清中の標的ウイルスは、予め抗体を修飾した磁気ナノ粒子に結合させて磁気分離を行い、不純物の影響を排除する。③磁場誘導による発熱で高分子ナノ小胞の複合体を溶解し、中に包まれた数千個のV2O5 NPを放出させ、触媒活性を飛躍的に向上させることで、比色法と電気化学的方法の2元的にウイルスの検出を行う。本研究は、ウイルス検出の最大の課題「不純物の影響と感度の向上」を解決するもので、既存のウイルス検出系の感度を遥かに凌駕する検出系の開発を目指すものである。

Outline of Annual Research Achievements

新規ナノ粒子包埋高分子ナノ小胞を作製し、高感度ウイルス検出に応用し、下記のような実績を得ている。
1)検出シグナルの増幅: V2O5ナノ粒子をカプセル化したリポソームを作製して、V2O5ナノ粒子のペルオキシダーゼ様活性と電気化学的酸化還元特性によるノロウイルスのデュアルモダリティセンシングプラットフォームを構築した。この方法で、高感度でウイルスの検出が可能であり、臨床ノロウイルスの検出も優れた精度で検出することができた。
2)量子ドット(QD)をウイルス様粒子(VLP)に組み込んだナノアーキテクチャの構築:in vitroで無機QDをE型肝炎ウイルス様粒子(HEV-LP)に詰め込み、HEV抗体検出用の蛍光バイオセンサーを開発した。HEV-LPを解体し、QDを組み込んだのち、自己組織化アプローチでHEV-LPを再構築した(QDs@HEV-LP)。ここに、抗IgGを修飾した磁気ナノ粒子、臨床検体、QDs@HEV-LPを混合するとQDs@HEV-LPに結合した。蛍光強度は、抗HEV抗体の濃度依存的であった。この方法で、HEVに感染したサルの感染後0日から68日間採集した血清から抗HEV抗体の診断に成功した。
3)高分子ナノカプセル:色原体、3,3'、5,5'-テトラメチルベンジジン(TMB)を、水溶液中での共沈に成功し、ポリ(ラクチド-co-グリコリド)(PLGA)にカプセル化した。ジメチルスルホキシド(DMSO)を添加すると、PLGAナノカプセルが溶解し、TMBが放出される。このナノカプセルをインフルエンザウイルス(IV)の検出に応用したところ、緩衝液や血清中の検出限界(LOD)がそれぞれ32.37 fg/mLおよび54.97 fg/mLであった。
以上の様に、ウイルス検出のボトルネックとなったシグナルの増幅に成功し、ウイルスを感度良く検出することができた。
