NLRP3インフラマソームは、遺伝性炎症疾患やメタボリックシンドロームなど、様々な疾患との関与が報告されている細胞内タンパク質複合体であるが、その調節機構については未だ不明な点が多い。そこで、これらの疾患に対する創薬ターゲットを見出し、より効果的に創薬を進めるために、ヒトの約2万種類のタンパク質アレイを用いて、NLRP3と相互作用する新規タンパク質の網羅的探索を行った。

| Academic Significance and Societal Importance of the Research Achievements |
NLRP3遺伝子の恒常的活性化変異はクリオピリン関連周期熱症候群という自己炎症疾患の原因となる。しかしながら、クリオピリン関連周期熱症候群の臨床症状を示しながら、NLRP3遺伝子に変異のみられない症例がある。本成果は、その原因として未知の相互作用分子の存在を示唆するものであり、NLRP3インフラマソームに対する新規の分子標的薬の開発につながる可能性を秘めている。引き続き、解析を進めていきたい。
