Outline of Research at the Start

本研究では、動物が道に迷い静止した際に、海馬の場所細胞が移動中の約10倍速の早送りモードで再活性化される「リプレイ」現象に着目し、そのリプレイと、そこで表現されるエピソード記憶の因果関係を解明することを目指す。それは、独自の電気生理学的記録法に光遺伝学を活用した神経刺激法を組み込むことで、エピソード記憶を想起するメカニズムを、従来不可能であった神経細胞レベルで実証することでもある。同時に本研究では、認知神経科学、情報工学、電気生理学、光遺伝学を融合し、神経細胞活動が表現するエピソードの記憶を操作する独創的な技術を開発する。

Outline of Annual Research Achievements

本研究では、海馬の場所細胞が再活性化される「リプレイ」現象に着目し、そこで表現されるエピソード記憶の想起過程を解明することを目指した。エピソード記憶の想起メカニズムを調べるためには、同一の外部環境下で複数のエピソードを経験させる課題を構築する必要があるが、そのために開発した再構成可能な迷路を活用し、「いつ、どこで、なにを、どのようにする」というエピソードにおいて重要な要素となる時間についての海馬情報表現や、明示的な時間認知と海馬の関与を調べた。その結果、海馬CA1が明示的な時間の保持に重要な役割を持つことが示唆され、またそれが海馬神経細胞の活動頻度により表現されていることが示唆された。この時間研究に関する先行研究については、Neurobiology of Learning and memory誌上で総説として報告した。また、再構成可能な迷路の活用法については、Journal of Visualized Experiments誌上で報告した。更に、これまでに神経細胞活動を無線記録する手法を開発し活用することで、海馬の神経細胞がゴール方向を表現することも明らかにしてきたが、そのような無線神経活動記録に関する総説をMicromachines誌上で報告した。エピソード記憶を理解する上で、海馬神経細胞がエピソードの区切りをどのように処理するのかを理解することは重要である。そこで、海馬の神経細胞活動がトレッドミルを走るイベントの区切りをどのように表現するのかを調べた結果、イベントの開始と終了に特に反応する神経細胞を同定することができた。加えて、海馬がそのようなエピソードの区切りを処理する上で重要となる目的地との関連についても調べた結果、目的地を変更するためにドーパミン作動性ニューロンから海馬への入力が重要であることもわかった。本年度は、これらの知見によりエピソード記憶の理解を進展させることができた。
