Outline of Research at the Start

本研究の全体構想は、水圏生態系の頂点に位置し、進化学的に独自の分類群に属する水棲哺乳類の細胞内受容体のシグナル伝達系を対象に、環境汚染物質による系の撹乱を先端技術によって解析し、種特異的な感受性を評価することである。さらにその知見を基に、種差の原因となる感受性規定因子の分子的な理解を目指す。

Outline of Annual Research Achievements

今年度は、バイカルアザラシ（Pusa sibirica）のエストロゲン受容体α およびβ（bsERα およびbsERβ）を介したビスフェノール類（BPs）および水酸化ポリ塩化ビフェニル類（OH-PCBs）の転写活性化能をin vitroおよびin silicoのアプローチで評価した。
in vitroレポーター遺伝子アッセイの結果、ほとんどのBPsとOH-PCBsがbsERアイソフォーム特異的なエストロゲン受容体アゴニスト活性を示した。ビスフェノールAFは、bsERαとbsERβの両方に対して、試験した11種のBPsの中で最も低いEC50を示した。4'-OH-CB50と4'-OH-CB30は、それぞれbsERαとbsERβに対して、試験した9種のOH-PCBの中で最も低いEC50を示した。4'-OH-CB107だけがbsERに影響を与えなかった。 4-（（4-イソプロポキシフェニル）-スルホニル）フェノール（BPS-monoP）、4'-OH-CB72、4'-OH-CB121は、bsERαのみに対して弱い転写活性化能を示した。in silicoドッキングシミュレーションは、これらの化学物質のbsERへの結合親和性を表し、in vitroでの転写活性化能の結果を一部説明した。化学構造記述子とin silicoドッキングデータの両方を説明変数として用いた機械学習モデルにより、in vitroの結果を予測することができた。これらのモデルから、化学的極性、ファンデルワールス表面積、架橋炭素の構造、フェノール性OH基の位置、特定のアミノ酸残基とのリガンド相互作用が、bsERを介した環境汚染物質のアゴニスト活性を説明する重要な変数であることが示唆された。
