Outline of Research at the Start

本研究は次の4本の柱から成る。①siRNAの送達システム開発における新たな律速段階を定量的・速度論的に解明する。②pDNA送達による遺伝子発現における律速段階(TC/TL)の分子機構を解明し、最高水準のpDNA送達システムを創製する。③がん免疫療法のブラックボックスを開き、個別化医療に対応したがん免疫ナノ治療法を確立する。④疾患細胞のMtへの遺伝子送達法を確立し、Mt遺伝子治療の道を拓く。

Outline of Final Research Achievements

We have been developing our own gene/nucleic acids delivery system and found important rate limiting steps. In this study, we challenged to identify molecular mechanisms of these rate limiting steps. In addition, we applied these delivery systems for mitochondrial gene delivery as well as for cancer immunotherapy. We succeeded to develop a novel nucleic acids delivery system to enhance antitumor effects via a mechanism of cancer immunotherapy. We also succeeded to develop a novel gene delivery system to mitochondria based on genome editing technology. These new technologies will contribute to enhancing clinical translation for nanomedicine.

Academic Significance and Societal Importance of the Research Achievements

我々は1999年薬剤分子設計学研究室を創設以来(19年間)、ウイルスベクターに匹敵する優れた発現効率を有し、かつ、安全性にも優れた人工遺伝子送達システムの開発を進めてきた。本研究により、ナノDDSによる核酸医薬・遺伝子治療におけるブラックボックスを分子レベルで解明することにより、がん免疫療法の薬効を飛躍的に促進することに成功した。さらに、疾患細胞のミトコンドリア (Mt) への遺伝子送達システムを開発することにも成功し、Mt遺伝子治療の臨床応用への基盤技術を確立することに成功した。これらの研究成果は、大学発の革新的核酸ナノ医薬品の創出に大きく貢献することが期待される。
