Outline of Research at the Start

ハビタブル惑星の探査対象として赤色矮星まわりの惑星が注目されており、そこでのバイオマーカーの観測が期待されている。バイオマーカーの1つである“レッドエッジ”は、光合成生物が光波長を吸収することによって起こる反射スペクトルであるが、実際に光合成生物が利用し得る光波長やその進化と対応した詳細な検討はされてこなかったのが問題である。本研究では、可視光利用型光合成から近赤外光利用型光合成への進化を実験的に再現・検証することで、吸収利用する波長帯の変化がどのように且つどれだけ速やかに起こるのかを明らかにすることを目指す。

Outline of Final Research Achievements

To understand the evolutionary process of photosynthesis utilizing near infrared light, we choose chlorophyllide oxidoreductase, COR, as a model system. In this study, we constructed combinatorial mutation libraries of the genes encoding COR subunits. The designed mutations included amino acid substitutions close to the predicted substrate binding site. We attempted to pack a mutated gene or a E. coli cell possessing a mutated gene into a micro-droplet by the microfluidics technique. We also tested the enzymatic assay conditions for COR in the micro droplet.

Academic Significance and Societal Importance of the Research Achievements

本研究で確立した、または今後確立する実験系を複合することで試験管内進化した近赤外光型CORを摘出して、その進化発生の元となるアミノ酸変化を同定することが今後の課題である。これによって長波長吸収型光合成への進化を実験的に証明することができ、地球と同様の光質環境変化が見込まれる系外惑星においても同様の進化が起きることを示唆する。生命居住惑星の探索におけるバイオシグニチャー選定の際に重要な知見を提供することが期待される。
