Outline of Research at the Start

石油に代わる再生可能エネルギーを効率的に生産できる基盤技術の開発は、人類の永続的繁栄のために必須である。シアノバクテリアは、アシルACP還元酵素（AAR）とアルデヒド脱ホルミル化オキシゲナーゼ（ADO）という2つの酵素を用い、光合成で軽油相当のアルカンを合成できることから、地球温暖化の防止に有効な再生可能バイオエネルギーの生産源として注目されている。しかしAARとADOの活性は低いため、両酵素の高活性化が急務である。そこで本研究では、実験と理論の両アプローチでAARとADOの酵素活性を飛躍的に向上させた変異体を創出した後、それらをシアノバクテリアに導入し、バイオ燃料の大量生産の実現を目指す。

Outline of Annual Research Achievements

シアノバクテリアは、アシルACP還元酵素（AAR）とアルデヒド脱ホルミル化オキシゲナーゼ（ADO）という2つの酵素を用い、光合成で軽油相当のアルカンを合成できることから、地球温暖化の防止に有効な再生可能バイオエネルギーの生産源として注目されている。しかし両酵素の活性は低いため、高活性化が必要である。そこで本研究では、複数の方法によってこれらの酵素活性を向上させ、微生物を用いたバイオ燃料生産を効率化させることを目指している。2022年度は次の研究を行った。
(1) 前年度までに得られたAARの様々なアミノ酸置換変異体のデータベースを用いて、複数のアミノ酸置換を組み合わせた多重変異体の構築を行った。
(2) 前年度に引き続き、タンパク質設計用ソフトウェアRosettaを用いてADOの酵素活性を向上させうる変異体の理論的設計を進めた。
(3) 細胞内で活性をもつ酵素量を増やすためには、AARとADOの可溶性を向上させる必要がある。そこで、AARとADOの末端に可溶性タグを付加するという合理的設計により、微生物細胞内での可溶性酵素量を増加させ、アルカン合成量を増大させることができた。
(4) タンパク質の機能を解明して改良するためには、タンパク質の立体構造を解析することも重要である。そこで、X線溶液散乱法を用いたタンパク質の立体構造解析も行った。また、細胞のような多分散系に存在する各タンパク質の大きさや形状をX線で解析する手法の開発を行った。
(5) 新規有用タンパク質を理論的に設計する際の成功率を向上させるために、Rosettaソフトウェアを用いた設計を様々なタンパク質に適用した。特に、アレルギーや白血病などに関わるタンパク質間相互作用を阻害しうるタンパク質やペプチドを理論的に設計し、実験で検証した。

Reason

本研究では、AARとADOの高活性化に向けて複数のアプローチを並行させて取り組んでいる。いずれのアプローチにおいても着実な進展がみられている。また、酵素に可溶性タグを付加するという合理的設計によってアルカン合成量を増大させることもできた。以上のことから、おおむね順調に進展していると判断される。

Strategy for Future Research Activity

今後は、これまでに行ってきた酵素高活性化の最終的な仕上げとして、得られた高活性化変異体の詳細な特徴づけや、それらの高活性化変異をさらに多重に組み合わせた変異体の構築などを行う。また、得られたAARとADOの高活性化変異体をシアノバクテリアに導入して、シアノバクテリアにおけるアルカン合成量の増大を目指す。
