Cyanobacteria can synthesize diesel-equivalent alkanes through photosynthesis using two enzymes, acyl-ACP reductase (AAR) and aldehyde deformylating oxygenase (ADO). However, since AAR and ADO have low activity, improving the activity of both enzymes is necessary to efficiently produce carbon-neutral renewable biofuels. In this study, we created highly active mutants of AAR and ADO using various approaches such as directed evolution experiments, mutational analysis experiments, and rational design. By introducing them into cyanobacteria, we succeeded in increasing the production of diesel-equivalent biofuels through photosynthesis.
