Outline of Final Research Achievements |
In this study, we focused on the characteristics of aromatic amino acid residues that are buried within the internal structure of proteins in their native state but exposed on the protein surface during the thermal denaturation process. We have developed chemical modification methods targeting tyrosine and histidine residues. The tyrosine residue-specific labeling reaction we developed specifically targets the exposed tyrosine residues on the protein surface. Our aim was to visualize the protein's thermal denaturation state, and for this purpose, we have developed a practical and efficient approach for modifying tyrosine residues in protein structures. Additionally, as a secondary outcome of this study, we successfully developed a method for modifying histidine residues in proteins.
