Outline of Research at the Start

本研究では、食中毒細菌によるリスク低減のため、バクテリオファージ(以降ファージ)およびファージの効果を促進させる安全性の高い天然成分を併用した効果的な食中毒細菌制御に寄与する学術的基盤を確立する。このため、腸管出血性大腸菌、サルモネラ、カンピロバクター、黄色ブドウ球菌、リステリアに対する溶菌ファージを単離してその性質を明らかにする。また、細菌の持つ免疫システム以外の溶菌ファージに対する耐性化に関与するシステムを解明し、ファージ耐性化を阻害する天然成分を植物抽出物などから見出してファージとの併用効果のメカニズムを解明する。

Outline of Final Research Achievements

Lytic phage against various food-poisoning bacteria including antibiotic-resistant bacteria have been successfully isolated from various foods, and elucidated their characteristics. The phages included the phages showing lytic activity even at low temperatures. From the Escherichia coli single gene-deficient strain library, 10 gene-deficient strains with higher phage sensitivity than wild-type strains were identified. Of these gene-encoded proteins involved in the phage resistance, myristin and chloramphenicol, which have been reported to inhibit the functions of DnaK and PriA proteins, respectively, increased the phage sensitivity of bacteria and suppressed the re-growth of resistant bacteria when used in combination with phages. The combined use with the food additive EDTA at 0.26 to1.3 mmol/L was also effective in improving phage sensitivity and suppressing the growth of resistant bacteria. These results will contribute to establish methods to control food-poisoning bacteria.

Academic Significance and Societal Importance of the Research Achievements

本研究の成果は以下の点で学術的、社会的な意義がある。海外で流行している食中毒細菌の菌株(遺伝子型や表現型)は、日本で食中毒を起こしている菌株とは異なるため、日本で問題となっている食中毒細菌の菌株に対して有効なファージを日本で分離して実用化する必要がある。今後、チルド流通による加熱殺菌後の高品質な食品のロングライフ化が期待されている。従って、低温保存中にも食中毒細菌を効率的に制御できるファージはチルド流通食品の安全性確保に必要である。ファージ感受性に関与する遺伝子の同定は細菌のファージ耐性化機構の抑制を可能とする学術的基盤となるもので食中毒細菌制御におけるファージ実用化のためには重要である。
